Cellular gene delivery via poly(hexamethylene biguanide)/pDNA self-assembled nanoparticles

Cellular gene delivery via polycations has wide implications for the potential of therapy, but it remained a challenge due to plethora pre- and post-uptake barriers that must be overcome reach desired efficiency. Herein we report poly(hexamethylene biguanide) (PHMB) as nano-vector intracellular plasmid DNA (pDNA) oligodeoxynucleotides (ODNs). PHMB pDNA or ODNs self-assembled into complex nanoparticles at different pH values (7.4 12). Their size, charge, cellular uptake, gene-expression efficiency are assessed compared PEI analogues. The systematic results show effective in delivering model cell lines culture (HepG2, HEK293T, HeLa), with measurable changes expression levels, comparable and, some conditions, even higher than PEI. well-accepted safety profile makes valuable candidate consideration an vector further study clinical translation.
